Effects of Adriamycin and Dimethylbenzanthracene on the Oxygen Toxicity in Mouse Brain

Abstract
We studied the changes of the activities of superoxide dismutase (EC 1. 15.-1. 1, SOD) and catalaes (EC 1. 11. 1. 6) and the production of superoxide radical in mace brain after the treat-went with adriamycin (ADM) and dimethylbenzanthracene (DMBA). ADM was injected subcutaneously (5 or 10 mg/kg body weight). After 16 days, DMBA (20 mg/kg) was applied on the skin once a week for 4 weeks.
The results were summarized as follows:
1. In all experimental groups, the activities of Cu, znSOD, MnSOD and catalase were lower than those in the controls (p< 0.01).
2. Production of superoxide :radical in all experimental groups increased in comparison with its control value (p< 0.01).
In conclusion, ADM and DMBA increased the superoxide radical production in mouse brains and decreased the activities of SOD and catalase, defence enzymes against oxygen toxicities. Therefore, oxygen toxicities could be developed by the treatment with ADM¢¥ and DMBA.
